摘要:目的 观察大鼠心包组织中C-kit, Sca-1, Nanog 阳性细胞的表达及分布,为治疗心肌损伤寻找新的干细胞来源。方法 取大鼠新鲜心包,利用石蜡切片和铺片技术,HE和Masson染色,观察其组织结构;利用免疫组织化学染色技术,观察C-kit、Scal-1、Nanog阳性细胞在心包组织中的表达;利用免疫荧光双标技术观察C-kit、Scal-1、Nanog之间的共表达情况;利用免疫印迹法检测心包组织中C-kit、Scal-1、Nanog的表达。 结果 大鼠心包组织中存在大量散在的小而圆或椭圆形细胞,广泛分布于成纤维细胞之间和小血管周围。免疫组织化学染色显示,这些细胞分别阳性表达C-kit、Sca-1、Nanog。免疫荧光双标染色显示,部分细胞共表达C-kit和scal-1两种表面抗原,部分细胞共表达scal-1和Nanog表面抗原,而C-kit与Nanog共表达于血管弹力膜。结论 大鼠心包组织中存在C-kit、Scal-1和Nanog阳性细胞,一种细胞可表达两种表面标记。

Abstract:Objective To investigate the expression and distribution of C-kit, scal-1 and Nanog positive cells in the rat pericardium in order to find a new source of stem cells for the treatment of myocardial injury. Methods Fresh rat pericardium was taken and prepared by paraffin section and surface spread technique, then the tissue structure was observed by HE and Masson staining. Immunohistochemical staining technique was used to observe the expression of C-kit, scal-1,and Nanog positive cells in the pericardium. The immunofluoscence double labeling staining technique was used to observe co-expression of scal-1, Nanog and C-kit proteins in pericardium. Western blotting technique was used to detect expression of C-kit, scal-1 and Nanog proteins in pericardium. Results A large number of small round (oval) cells were observed in the pericardial tissue of rats, which scattered among the fibroblasts and around the small vessels. Immunohistochemistry showed that these small round cells positively expressed C-kit, Sca-1 and Nanog proteins. Immunofluorescence double-labeled staining showed that some cells co-expressed both C-kit and scal-1 surface antigen. Some cells co-expressed scal-1 and Nanog, while C-kit and Nanog co-expressed in vascular elastic membrane. Conclusion There are C-kit, scal-1, Nanog positive cells in the rat pericardium tissues, and one kind of cells can simultaneously express two kinds of surface antigen markers.
